Overview
Muk has massive HP and Attack, and has the strongest Poison attack in the game, Sludge, among other moves. Still though, Muk is Poison-typed, which is arguably the worst type in the game. One of the nice things about Muk though is having the third most destructive attack in the game, his Explosion (under Golem's and Snorlax's Self-KO moves), and can take out many Pokemon. Muk is slow though and his high HP gets drained pretty quickly in an environment full of Psychic and Ground attacks.

Sludge is Muk's main damaging move, while Explosion is his finishing move. Acid Armor and Mega Drain are almost entirely for Golem and Rhydon, which actually put the match-up in Muk's favor. Other possibilities include Screech to support Sludge and Explosion, Body Slam for Paralysis and Thunderbolt for defensive aquatics like Articuno and Cloyster. In general though, this moveset is fine on its own.
The problem with Sludge being Muk's main attacking move is that it causes Poison and is the worst offensive type. By Poisoning opponents, Muk is only giving them immunity to other (better) status conditions, and Sludge has low base power too. On top, Sludge is only super-effective against six final form Pokemon and only one (Exeggutor) is commonly used in OU. As bad as it is though, it is his strongest damaging move outside Hyper Beam and Explosion. Next, while his Explosion is lethal, it's very obvious, so the opponent will know to be ready for it, either using a Rock, Ghost, defensive Pokemon or something with Reflect, etc. And also, while Screech helps support his main moves, Muk can't really cause pressure with switches except with Explosion, so it's very easy to deal with.
Muk's main use is trying to use Explosion on something important, such as Chansey or Alakazam. While Snorlax does this better, Muk at least has different weaknesses than Golem (particularly Ice), so Muk can actually be usable and not just replaced by something else (though Snorlax is better in almost all cases). Acid Armor and Mega Drain are enough to beat Golem and Rhydon, barring a Critical Hit, but Muk is helpless against Gengar. So overall, Muk does require a little bit of setting up and prediction to be used effectively, and at most, Muk will usually just take out one Pokemon.
Counters
Gengar is by far the safest choice, easily taking anything Muk can do. Golem and Rhydon can also work if Muk is Paralyzed and for jumping in on Explosion. Usually though, you want to take Muk on defensively and be ready for Explosion. There is no attack that can OHKO Muk without a Critical Hit, so Muk can use Explosion immediately. Alakazam with Reflect, Starmie and other defensive Pokemon are fine. Focus more on handling Explosion as Sludge doesn't need to be worried about too much and you may actually want to try to get Poisoned if possible for immunity to other status conditions. Don't be afraid with obvious baits against Muk as the player needs to be playing perfectly to keep up, and often only does small damage with Sludge.
